新規バイオマーカーであるオートタキシンの臨床的有用性

はじめに

　オートタキシン（autotaxin：ATX）は血漿中に存在するリゾホスホリパーゼDである．細胞運動促進作用をはじめとする，多彩な生理活性を有するリゾホスファチジン酸（lysophosphatidic acid：LPA）産生における重要な酵素である．もともとは1992年にヒト悪性黒色腫株であるA2058の培養上清より単離された糖蛋白質であり，当初より腫瘍細胞の運動を促進させる作用をもつ因子として，癌の転移や浸潤に関与する可能性が示唆されていた．さらにATXがもつLPA産生能が明らかになったことにより，癌や組織の線維化，血小板凝集などとの関連性についても，生体内での機能解析が急速に進められることとなった．現在，ATXはさまざまな疾患のバイオマーカーとしての役割の他，疾患の治療標的としても注目されている．本稿では，肝線維化を中心に，これまで明らかになっているATXの臨床的有用性について述べる．
