加齢関連疾患に対する選択的老化細胞除去というアプローチ

はじめに

 加齢や加齢性疾患に伴い,全身の臓器で老化細胞が蓄積する.これらの細胞が臓器の機能的・構造的リモデリングを進展させることを示すエビデンスが集積されている.老化細胞は老化関連分泌表現型(senescence-associated secretory phenotype:SASP)を介して慢性炎症を惹起し,動脈硬化性疾患,心不全,非アルコール性脂肪性肝炎,慢性腎臓病などの加齢性疾患の病態を増悪させる.しかし,細胞老化は無秩序な細胞分裂を抑制する役割も担うため,細胞老化を抑制することでがん化の懸念が生じる.

 そこで今日,“選択的老化細胞除去”というアプローチが注目されている.本稿では,老化細胞を標的とした治療,特に選択的老化細胞除去薬について解説する.
