アレルギー疾患のトランスクリプトーム解析Ⅰ

1. はじめに

　周知のように個体(＝細胞)がもつすべての遺伝情報の意味であるゲノム(genome)は遺伝子(gene)と染色体(chromosome)を合成した用語である．一方，トランスクリプトーム(transcriptome)という用語は，1つの細胞に存在するすべてのmRNA(transcript)情報のことを指す(図1)．ヒトゲノム配列のほぼ全容が明らかになり，一度に数万のmRNAを定量できる装置であるマイクロアレイが開発された前世紀末から登場した新用語である．当初は解析手法であるDifferential display PCRやマイクロアレイの技術が未発達なこともあり，プロテオーム(proteome)という用語のほうがより知られていたが，ここ数年のマイクロアレイ技術の技術的進歩はめざましく，今や多くの研究施設において，PCR法やフローサイトメーターなどと同様に日常研究レベルで全遺伝子のmRNAへの発現状態を定量することが行われるようになっている¹⁾．

　本稿においてはアレルギー疾患病態解析のトランスクリプトーム解析の現況について筆者らの研究を中心に紹介させていただくことにする．
