1. はじめに

　生体内の蛋白質のうち約50％は糖鎖が結合した糖蛋白質であるといわれている．糖蛋白質中の糖鎖は，発生，分化，癌転移，蛋白質の輸送機構，老化，ホルモンの活性調節など広く生命現象に関与することが明らかになりつつある．また，糖鎖は炎症，リウマチ，肝硬変などの種々の疾患や癌化に伴い特徴的な構造変化を起こすことが明らかにされつつあり，診断マーカーとしても糖鎖は注目されつつある．

　癌や炎症などの様々な病態時に糖蛋白質の糖鎖が質的あるいは量的に変化することが注目され，これらを解析するための技術も目覚しく進歩した．なかでも，キャピラリー電気泳動法(CE)は現在最も高感度かつ高分解能分離が期待できる分析法であり，紫外部吸収検出だけでなく，レーザー励起蛍光検出法を組み合わせることにより10-¹⁵～10-¹⁸molという超微量の糖鎖を検出することができるため，生体組織や血清などから得られた微量の複合糖質試料中の糖鎖解析にも対応できる．

　本稿ではCEを利用する糖鎖解析について，蛋白質より遊離したN結合型糖鎖を解析する方法と，糖鎖構造の違いに基づく蛋白質のマルチフォームであるグライコフォームを糖鎖構造の違いに基づき直接解析する方法の臨床応用について解説する．また，CEの高速化を目指したマイクロチップ電気泳動によるグリコサミノグリカン類の簡易分析法についても紹介する．
