はじめに

　メタボリックシンドローム(metabolic syndrome；MS)は，生活習慣(すなわち環境因子)と複数の遺伝因子がその発症に関与する，いわゆる複合遺伝形質(complex trait)と考えられる．代表的な環境因子としては，摂取総カロリーのみならず動物性脂肪や単純糖質(ショ糖など)の摂取過多，食物繊維やビタミンの摂取不足など食事内容に関するものがよく知られている．加えて，運動不足やそれに伴う肥満，不規則な生活サイクルに起因する自律神経系のアンバランスなどが指摘されている．一方，関与する遺伝因子の解明はいまだ不十分にとどまっているが，ヒトゲノムプロジェクトの完成，それに続くHapMap計画の進展，さらにはハイスループットな解析技術の登場により，近年興味深い進展がみられている．本稿ではMSにおける脂質代謝異常の分子機構を概説したあと，MSの遺伝因子についての最近の知見を脂質代謝異常の観点からまとめてみる．
