グライコミクスの今後と検査とのかかわり

はじめに

　最新医学講座，「糖鎖と臨床検査シリーズ」は本号で終了する．この間にも糖鎖に関する研究論文や総説が多数報告されておりこの分野の研究が注目されていることを示している．本シリーズも執筆された各専門の先生方のわかりやすい教育的な総説を拝読して，日本の糖鎖研究の進歩性と層の厚さを実感されたことと思う．

　しかし指摘されているように複雑な分子構造を持つ糖鎖は，遺伝子のように短時間で解析が終わることはありえない．個々の糖鎖について病態生理機能の解析など，各分野の研究は着々と進んでいるが，なお不明確な点が多い．本質的なところでは糖鎖修飾後を糖蛋白分子の翻訳生成物とすると，糖鎖はどのような形で発現調節にかかわっているのか．癌などで同じアポ蛋白分子に対し，糖鎖変化が生じるのはなぜかなど，知りたいところである．

　最近，糖鎖は特異的な分子間認識や接着などに加え，糖鎖の新しい機能として，シグナル伝達分子としての側面を持つことが明らかになってきた．その一つは図1に示すように，核内蛋白のスレオニンやセリンのリン酸化サイトに糖のβ-N-アセチルグルコサミン(β-N-acetylglucosamine)が競争反応的にO-型結合反応¹⁾をし，蛋白の発現や輸送に直接関係しているとの報告である．表1のようにこの糖鎖の乱れがいくつかの疾病を引き起こすことがわかる．

　検体検査の分野でも糖鎖は癌，感染症関連のマーカーなど，すでに古くからかかわりをもっている．しかし最近の基礎分野における急速な研究成果の蓄積を考えると，少し注意が不足していたように思われる．本稿ではグライコミクスの今後に対する2，3の期待を述べるとともに，近い将来，臨床検査にかかわりを持ちそうな糖鎖の研究分野を予測し，取り上げてみた．
