クロマチン構造変換因子と癌―Brm型SWI/SNF複合体と胃癌

1．はじめに

　ヒトゲノム計画が終了し全遺伝子配列を入手可能になった昨今，生物学・医学の理解にエピジェネティクスの概念が不可欠であることが，クローズアップされるようになった．“one-hit/two-hit theory”に代表されるように，これまでジェネティクスの異常が引き起こす代表的疾患と考えられてきた癌(悪性腫瘍)も例外ではなく，近年，遺伝的変化を伴わずに癌の発生・進展に影響を与える機構が次々と明らかにされてきている．エピジェネティクスの全貌は広汎にわたるが，現時点では，「メチル化」「ヒストン修飾」「クロマチン構造変換」が3本柱であると考えられている．

　本稿ではヒト細胞中のクロマチン構造変換因子の中で特に重要と考えられているSWI/SNF複合体に焦点を当てて概説し，胃癌との関連を発見したわれわれの最新の知見を紹介したい．
