文献購入ページに移動 遺伝子検索はpolymerase chain reaction(PCR)の普及により身近なものとなってきた.しかしながら,thermal cyclerを用い2~3時間が必要で,迅速性に問題がある.60分間以内で結果の出るreal time PCR法も開発され遺伝子数の定量的検索も可能であるが,高価な機器が必要であり,ベッドサイドへの応用には至っていない.近年わが国で開発されたloop-mediated isothermal amplification(LAMP)法は独創的なprimerを設定し2本鎖をはがしながらDNA合成を進めるBst polymeraseを用いて60~65℃の一定温度で反応を進め,1時間以内で結果を得ることが可能である.LAMP法は特異性,感度も高く感染症の遺伝子レベルでの迅速診断の可能性を持つ発展性の高い検査法と考えられる.LAMP法の臨床応用を解説する.
