1.はじめに

 近年,リゾリン脂質メディエーターであるリゾホスファチジン酸(lysophosphatidic acid;LPA)とスフィンゴシン-1-リン酸(sphingosine-1-phosphate;S1P)の多彩な生理活性と生体内における役割の解明は飛躍的に進んだ1,2).LPA,S1Pについて特徴的である点は,他の生体内メディエーターに比して血中濃度が高く,in vitroの細胞系で作用を及ぼす濃度に非常に近いということである.すなわち,メディエーターとして多彩な作用を有し,血中でも作用を及ぼす可能性が高いレベルで豊富に存在するため,受容体発現量の変化により,その作用のオンオフが調節されている可能性も推定され,実際に受容体アゴニスト,アンタゴニストが疾患治療に応用されつつある.

 いずれにせよ,LPA,S1Pともに様々な疾患への関与が報告されてきており,本稿では,特に肝臓疾患における役割についての報告を概観する.
