コンドロイチナーゼABC(C-ABC)―椎間板ヘルニア,脊髄損傷治療への応用

■はじめに

 コンドロイチナーゼABC(C-ABC)は,グラム陰性桿菌の一種であるProteus vulgarisから分離・精製された酵素29)であり,コンドロイチン硫酸などのグリコサミノグリカンを分解する活性を有している.グリコサミノグリカンは,コアプロテインに共有結合してプロテオグリカンの形をとり,コンドロイチン硫酸プロテオグリカン(CSPG)などと呼ばれる.ある種のCSPGのコアプロテインは,ヒアルロナンに結合して巨大な会合体を形成し,その著しく高度な親水性のために分子中に多くの水分子を保持する.その性質が椎間板内髄核の圧縮的衝撃力の吸収作用の基となっている.C-ABCは髄核のグリコサミノグリカンを分解することによってその高い保水力を減弱させうることから,椎間板ヘルニアにおいて椎間板内圧を低下させる目的で臨床応用が期待されている.また,CSPGは神経軸索の伸展阻害効果があり,損傷後の脊髄に強く発現している.C-ABCはそのCSPGを分解することにより,損傷脊髄において軸索再生を促進する目的で脊髄損傷治療への応用が期待されている.
