プロテオミクス解析により同定された広義のアポリポ蛋白

　ポストゲノム時代で注目されているプロテオミクスは，検体の複雑な前処理なしでハイスループットに解析できるMALDI-TOF-MS，SELDI-TOF-MSなどの質量分析により，従来の二次元ポリアクリルアミドゲル電気泳動(2D-PAGE)レベルでは解析が困難であった低分子蛋白質やペプチドの検出が可能になったことで，疾患・病態のさらなる詳細な評価を可能にした．従来アポ蛋白は，リポ蛋白の組織・細胞への作用にかかわる認識シグナルとして機能する構造蛋白として位置づけられていたが，そのほかにリポ蛋白粒子表面には多くの結合蛋白が局在しており，脂質代謝のみならず，血栓，炎症，酸化還元など多岐にわたる機能を発揮している．最近のプロテオミクス研究の成果から，従来のアポ蛋白以外のリポ蛋白結合蛋白やペプチドが発見され，LDLやHDLの亜分画が特徴的な機能を発揮するユニークなプロファイルをもつことが確認されている．LDLやHDLに結合する蛋白のプロテオミクスから，新たに信頼されるバイオマーカーの確立や量と質を兼ねて評価できるリポ蛋白関連検査の開発はこれからの動脈硬化性疾患などの診療や予防に有用であるが，それには高い精度と簡便性を備えたプロテオミクスへの発展が重要な鍵となる．
