1.はじめに

 リポ蛋白による末梢-肝臓間のコレステロール輸送は古くから確立されており,血中LDLコレステロール(low-density lipoprotein cholesterol),HDLコレステロール(high-density lipoprotein cholesterol)といった血中リポ蛋白の解析が動脈硬化の指標として利用されている.近年では,このリポ蛋白の研究はより細分化されてきており,酸化リポ蛋白,リポ蛋白酵素,リン脂質など,様々な方面から解析されている.このリン脂質は特に,ここ数十年に飛躍的に発展してきた物質であり,単なる構造膜や中間代謝産物ではなく,シグナル伝達物質として,多彩な生理活性作用を発揮することが明らかになってきた.

 スフィンゴシン1-リン酸(Sph-1-P)は,グリセロリン脂質であるリゾホスファチジン酸とともに多大な注目を浴びている脂質性メディエーターであり,最近の研究で,HDLのコレステロール輸送以外の抗動脈硬化作用〔一酸化窒素(NO)産生,遊走,増殖,細胞生存,接着分子発現抑制といった血管内皮細胞の保護的作用〕の一端を担っていることもわかってきた.

 本稿では,最近の話題として,このSph-1-Pの概要,および,Sph-1-Pと動脈硬化とのかかわりについて述べたい.
