長鎖非コードRNAの性状と疾患への関与

トランスクリプトーム解析によってヒトゲノムの9割以上の領域から，正体不明の非コードRNAが生み出されていることが明らかにされた．近年これらの機能解析が進み，非コードRNAが重要な遺伝子発現制御因子として機能することが明らかになってきた．特に非コードRNAが，エピゲノム制御のヒストン修飾酵素の標的特異性を規定するというモデルが提唱され，実際ある種の癌細胞で，非コードRNAの著しい発現上昇に伴い一群の遺伝子近傍のヒストン修飾パターンが変化することも確認された．一方で，難治性神経変性疾患の原因蛋白質と特異的な非コードRNAとの結合が，患者脳で著しく上昇する例も見出された．さらには疾患細胞で発現変動する非コードRNAも続々と見つかっている．本稿では，非コードRNAと疾患との関連についての研究背景と解析法について解説する．
